Genetic diversity analysis using random amplified polymorphic DNA (RAPD) markers in mungbean (Vignaradiata L. wilczek)

Abstract

Total 28 mungbean genotypes were used for studying genetic diversity using 15 Random Amplified Polymorphic DNA (RAPD) primers. All primers produced polymorphic amplification products. In all 1027 amplification products were generated with on an average 68.46 bands per primer. The fragments produced from all 28 genotypes varied widely in size from 120 to 2500 bp. The dendrogram resolved all genotypes into five major clusters viz., 1, 5, 5, 16 and 1 genotypes respectively. Cluster IV was classified into 5 sub clusters Ato E with 1, 4, 1, 4, and 8 genotypes respectively. However, the dendrogram did not showed any significant correlation between the genetic divergence and geographical distribution. Squared Euclidean distances ranged from 3 to 59, indicating diversity among 28 genotypes related to their performance in field conditions. PBM and BL 849, resistant to mung bean yellow mosaic virus (MYMV) were found to be distinct for seed yield per plant, pod yield per plant, test weight, seeds per pod, pod length, number of pods per plant, clusters per plant, number of primary branches per plant, plant height. Hence, they can be used in breeding programme for their desirable characters.
