Direct organogenesis in Dianthus caryophyllus Linn

Abstract

Adventitious shoots of carnation (Dainthus coryphylus Linn.) were regenerated via direct organogenesis from shoot-tip and nodal segment of in vitro raised shoots by culturingthem on MS medium supplemented with various combinations at different concentrations of plant growth regulators. The optimum medium for shoot induction from the shoot tip as well as nodal segment was found to be MS medium supplemented with 0.5 gm/1NAA, 0.5 gm/1 Kinetin and 0.5 mg/1 GA3. This medium produced normal shoots with no signs of hyperhydricity. The maximum rooting response from the in vitro raised shoots was found on MS medium supplemented with 0.5 mg/1 IbA. Among the different carriers tested for successful transfer of in vitro raised plantlets, higher survival rate of 90% was recorded in the carrier vermiculite followed by cocopeat (85%).
