Abstract
Objective: To study the expression of IFN-γ and IL-6 in the peripheral blood mononuclear cells of systemic lupus erythematosus (SLE) patients with pathogensis at single-level. Methods: Peripheral blood mononuclear cells (PBMC) from 20 patients with SLE stimulated for 6 hours with phorbol myristate (PMA) plus ionomycin in the presence of monensin. Cells were stained for intracellular cytokines such as IFN-γ and IL-6 by enzyme linked immunospot assay (ELISPOT). Results: The expression of IL-6 and Th2/Th1 ratio were significantly increased in patients compared with 10 healthy individuals (P<0.01). Conclusion: The increased expression of IL-6 and a shift toward Th2 Cytokine profile is observed in the PBMC of SLE patients. Th1/Th2 cytokine balance has important implication in the pathogensis.
