Abstract

A B S T R A C T The goal of this study is to establish protocol for callus induction from cotyledon leaf explant from in vitro microplant of Callotropis prossera using two types of auxins 2, 4-D (2, 4- dichlorophenoxy acetic acid) and NAA (Naphthalene acetic acid), Calotropis procera. Murashige and Skoog ( MS) medium verified with 1.5 mg/l 2, 4 achieved 100% of callus after ten day, 2,4-D at all concentration achieved 100% of callus. NAA display varied percentage of callus (4 5%-75%). The highest callus degree was (3.75±0.1) obtained on the MS medium supplemented with 1.5 mg/l 2,4-D,followed by (3.05±0.2), obtained on MS media supplemented with 2.0 mg/l 2,4-D,the texture of callus is watery and color is creamy. Maximum callus induce by NAAwas (1.00 ±0.2) obtained on MS media supplemented with 1.5 mg/l NAA. There was significant variation on callus response between the two hormones, on the callus percentage, callus color, degree of callus. The auxin2.4 D induce watery creamy callus with high percentage and degree of callus response, while NAA induce friable green callus with low callus percentage and degree for callus response.
