Callus induction and shoot regeneration in strawberry (Fragaria x ananassa Duch.)

Abstract

Effect of different growth regulators on in vitro responses of strawberry for callus induction and shoot regeneration has been investigated. Variable callus was induced in strawberry depending upon the types of explant and combinations of plant growth regulators. Leaf disc of mature leaf inoculated in MS medium supplemented with 1.0 mg/l 2,4-dichlorophenoxy acetic acid+ 0.1 mg/l 6-benzylaminopurine was found to be the best for high frequency callus induction. Plant regeneration from callus was also influenced by source of explants and used growth regulators. The highest number of shoot per inoculum was obtained in MS medium suplimented with 2.0 mg/l 6-benzylaminopurine + 0.5 mg/l α-napthalene acetic acid. In shoot regeneration mature leaf derived callus gave the best result followed by young leaf and internode derived callus. After successful shoot regeneration, regenerated shoots were cultured for root induction in MSo. The developed plantlets through in vitro culture technique were acclimated and successfully transferred to in vivo condition for further evaluations.
