Abstract
Theincorporation ofhyperimmune serumintocell culture mediumtocontrol endogenous viral infections ofprimary cells canhaveasignificant effect onthe replication ofother viruses. Whencommercial simian virus 5orsimian virus 40 antiserum wasusedwithprimary monkeykidney cell cultures, we founda significant inhibition (>90%) ofthereplication ofparainfluenza virus types2and 3andreovirus type1.Intheviral diagnostic laboratory, theuseofhyperimmune serumwithprimary monkeykidney cells mayresult infailure toisolate certain viruses ifthese cells arenotfirst washedfree ofhyperimmune serum. Simian virus 40(SV40) isendogenous toseveralspecies ofAsianmacaques andwasfirst discovered incell cultures prepared fromrhesus monkeykidneys (17). Withthewidespread use ofprimary monkeykidney cell cultures, ithas becomecommonpractice toincorporate antiserumintoculture mediumtohelpcontrol simian viruses whichoften makecell cultures unsuitableforviral isolations. Forexample, ithasbeen recommended that0.2to1.0%hyperimmune simian virus serumbeusedtocontrol certain simian virus infections (3-5), andnowitiscommonforcommercial biological suppliers toadd simian virus antiserum toprimary monkeykidneycells. Theincorporation ofhyperimmune serumintocell culture mediumcould affect the isolation andgrowth ofother viruses. Thepurposeofthis report istodemonstrate thatcertain commercial lots ofSV5andSV40antisera incorporated intocell culture mediumhaveaninhibitory effect onthereplication ofparainfluenza virus types2 and3,reovirus type1,andthe simian rotavirus, SA11.
