Abstract
Objective Uo study the possibility of the human papillomavirus type 58 early gene E2 as a candidate antigen of DNA vaccine.Methods Recombinant eukaryotic vector pcDNA3/58E2 was constructed and was transfected into SiHa cells.HPV58 E2 mRNA transcripts were examined by RT-PCR.BALB/C mice were immunized by muscle injection with plasmid pcDNA3/58E2,and serum specific anti-HPV58E2 IgG was detected by ELISA.Results HPV58 E2 mRNA transcripts were detectable in SiHa cells by RT-PCR,and specific anti-HPV58E2 IgG could be induced in mice immunized with plasmid pcDNA3/58E2.Conclusion The recombinant plasmid pcDNA3/58 E2 was successfully constructed,and it could stimulate an E2 specific immune response in mice as a DNA vaccine.
