Abstract
A modified Masson's trichrome method for use on plastic-embedded tissues is presented. The trichrome stain can be vastly improved by extending the iron hematoxylin, Biebrich scarlet-acid fuchsin, and aniline blue staining times. After rinsing in acidified water the blue-colored background can be removed by dipping in 2-butoxyethanol. This method gives good results on tissues that are embedded in glycol-methacrylate plastics which are not removed prior to staining. The effects are similar to those obtained with routine, paraffin-embedded tissue.
