Tissue Culture Media Optimization and Genetic Transformation of Jatropha curcas Genotype Jatromas Cotyledon Explants

Abstract

Jatropha curcas oil seed is used as an alternative energy to replace fossil fuel. J. curcas genome can be modified by adding desired gene using Agrobacterium transformation to develop J. curcas plant quality. The explants regeneration was carried out in tissue culture solid medium. However, established tissue culture method may not give the same result due to genetic variation from the explants. The media optimization was done by combining IBA and BAP for callus induction and combination of IBA, BAP, and GA 3 for shoot induction. The most optimum media for Jatromas callus induction was BIL and SIG for shoot induction.
