Abstract
An attempt was made to define factors important in choice of labelling conditions for thymidine incorporation into DNA as a means of quantitatively assessing DNA synthesis in human lymphocyte cultures stimulated by PHA and PPD. The effect of adherent cells on attaining and maintaining optimal labelling conditions was studied. It was found that the presence of adherent cells in PHA-stimulated cultures strongly affects requirements for exogenous thymidine, but has no effect in PPD-stimulated cultures.
