Dye-coupling in Tobacco Mesophyll Cells Surrounding Growing Tobacco Mosaic Tobamovirus-induced Local Lesions

Abstract

Some factors related to cessation of the movement of tobacco mosaic tobamovirus (TMV) in the late phase of a defence response were examined. Mesophyll cells surrounding the local lesions induced by TMV in N. tabacum cv. Xanthi-nc were micro-injected with fluorescent dye 2, 3 and 7 days post-inoculation. At 7 days post-inoculation, twelve out of 20 injections into cells adjacent to the lesion, showed the expected dye-coupling (outflow of fluorescent dye from injected cell to adjacent ones via plasmodesmata) whereas 17–20 out of 20 injections were successful in other cases. Callose inhibitor (tunicamycin), dark treatment and incubation of plants with ascorbic acid, which play a role in blocking plasmodesmata or induction of defence responses, did not seem to have an effect on lesion growth. These data imply that defects in the plasmodesmal function, although not total, may account for the formation of late local defence reaction together with other factors that restrict viral spread in the continuous cell-to-cell mode in mesophyll tissue. Zusammenfassung Untersucht wurden einige Faktoren, die mit dem Stop der Bewegung des Tabakmosaik-Tobamovirus (TMV) in der spaten Phase einer Abwehrreaktion in Verbindung stehen. Mesophyllzellen, die die Lokallasionen umgeben, welche durch TMV bei N. tabacum cv. Xanthi-nc hervorgerufen worden waren, erhielten 2, 3 und 7 Tage nach der Inokulation (d p.i.) Mikroinjektionen mit fluoreszierendem Farbstoff. Zwolf von 20 am 7. d p.i. ausgefuhrten Injektionen in Zellen neben Lasionen bewirkten die erwartete Farbstoffbindung (Ausfliesen fluoreszierenden Farbstoffs von injizierten Zellen in die benachbarten Zellen durch Plasmodesmen). In anderen Fallen waren 17 bis 20 von 20 Infektionen erfolgreich. Der Kallosehemmer Tunicamycin, Dunkelheit und eine Inkubation der Pflanzen mit Ascorbinsaure, die eine Rolle bei der Blockierung von Plasmodesmen oder der Induktion von Abwehrreaktionen spielt, hatten keinen feststellbaren Einflus auf das Wachstum der Lasionen. Diese Ergebnisse zeigen, das Defekte in der Funktion der Plasmodesmen gemeinsam mit anderen Faktoren, die die kontinuierliche Ausbreitung des Virus von Zelle zu Zelle in Mesophyllgewebe begrenzen, fur die Auslosung einer spaten lokalen Abwehrreaktion verantwortlich sein konnen. Dies gilt auch dann, wenn die Funktion der Plasmodesmen nicht vollstandig zum Erliegen kommt.
