Bacterial conjugation between pseudomonads in the rhizosphere of wheat

Abstract

Transfer of plasmid RP4 between introduced pseudomonads was studied in rhizosphere and non-rhizosphere soil of wheat, in soil chambers and in culture tubes. In both experiments, the presence of growing wheat roots stimulated the occurrence of plasmid transfers in the soil. The plasmid transfer frequencies in rhizosphere soil in the soil chambers were consistently higher than those in rhizosphere soil in the culture tubes, indicating an influence of the experimental set-up. In the soil chambers, both the survival of introduced donor and recipient strains and the plasmid transfer frequencies decreased drastically at increasing distances from the roots. In addition, plasmid transfer frequencies were influenced by the inoculum densities of both donor and recipient strains; higher frequencies were observed in soil that was initially inoculated with higher cell numbers.
