Abstract The identification ofNOD2(now calledCARD15) as a susceptibility gene for Crohn's disease was a major milestone in the genetics of inflammatory bowel disease, and appears to provide an important link between pathogen recognition and activation of the inflammatory response. Regions of extensive linkage disequilibrium and modest genetic effects are likely to pose significant challenges in the search for novel susceptibility genes.
