Abstract
This review presents a chronological discussion of the discovery of opiate receptors and the endogenous opioid peptides. After a summary of the properties and distribution of the receptors and their endogenous ligands, the possible function of the endogenous opioid system is discussed. Evidence for a role of this system in endogenous pain modulation and in opiate addiction is presented. The nature of the evidence and the absence of definitive proof of a causal involvement of the opiate receptor-endorphin system are emphasized.
