Evaluation of neutrophil VCS parameters by the hematology analyzer

Abstract

Th e increasin g amoun t o f immatur e an d ban d for m neutrophil s i n bloo d circulatio n are helpful in the diagnostic of inflammation or bacterial infection. Immature cells tend to be larger and have lower nuclear complexity than mature cells. Thus, the changing of mean volume and mean complexity of cells indicate the presence of immature cells in th e c ir c ul a ti o n . Th e C o ult e r L H 75 0 h e m a t o l og y a n a ly ze r u se s th e v o lum e (V) , co ndu c tivit y (C), and scatter (S) technology for ifferential white blood cell count. In this study, we evaluated the neutrophil VCS parameters, there were mean neutrophil volume (MNV), mean neutrophil conductivity (MNC) and mean neutrophil scatter (MNS) of K 2 ED T A blood samples, collected from 160 healthy subjects, who had normal hematologic data, with mean age 42 ± 13 years, the second group of VCS data was retrospectively analyzed from 700 samples who had normal hematological values and the last group, abnormal matologic data group was performed on 72 leftover blood samples with submitted to routine amination and in blood smear contained band form neutrophil. The results of MNV, MNC and MNS from healthy subjects were 142.0 ± 5.9, 147. 1 ± 3. 7 an d 144. 4 ± 6.9 , r espec ti ve l y . T h e V C S pa r ame t e r s o f r e tr ospec ti v e data were onsistent with healthy group. Patient group had significant higher MNV (150.7 ± 11.2, p < 0.0 0 1) and lower MNC (142.7 ± 4.7, p < 0.001), MNS (137.9 ± 10.3, p < 0.001) than healthy group. There were correlations between MNV and manual band form neutrophil count (r = 0.482, p < 0.001) and absolute neutrophil count (r = 0.328, p < 0.001 ) . H o w e v e r , m o r e sa mpl e s fr o m b ac t e ri a l inf ec ti o n a n d o th e r ca u se s o f n e utr o phili a are needed for further study to confirm the clinical usefulness of this preliminary data .
