Abstract

Sumatriptan (Imitrex®) is a selective 5-HT1B/1D agonist and was the first specific antimigraine medication. Since the introduction of sumatriptan, several second-generation triptans have entered the marketplace, including zolmitriptan (Zomig®), naratriptan (Amerge®), rizatriptan (Maxalt®), almotriptan (Axert®), frovatriptan (Frova®) and eletriptan (Relpax®). Their pharmacology and pharmacokinetics are presented briefly in the background section. The subsequent sections describe in detail the various syntheses of sumatriptan, zolmitriptan, naratriptan, rizatriptan, almotriptan, frovatriptan and eletriptan.
