Abstract

Objective:To investigate the effects of resveratrol(Res) on cell proliferation and apoptosis of gastric adenocarcinoma SGC-7901 cells and their action mechanism.Methods:SGC-7901 cells were treated with different concentrations of Res for various periods.The cell proliferation was examined by MTT assay.TUNEL method and flow cytometry were used to analyze cell cycle and detect apoptosis.Expressions of bcl-2,bax,caspase-3 and Fas proteins in gastric cancer cells were measured by immunohistochemistry.RT-PCR was used to detect the mRNA transcriptions of bcl-2,bax,caspase-3 and Fas.Results: Res inhibited the growth of gastric adenocarcinoma SGC-7901 cells in time-and concentration-dependent manners(P0.01,P0.05).Res induced apoptosis of SGC-7901 cells(P0.01).Res(150 μmol/L) significantly up-regulated the mRNA and protein expressions of bax,caspase-3,and Fas and down-regulated gene transcription and protein expression of bcl-2.Conclusion: Res inhibited the growth and induced apoptosis of gastric adenocarcinoma SGC-7901 cells in time-and concentration-dependent manners.It was related with up-regulation of expression of Fas,bax,caspase-3 and down-regulation of expression of bcl-2.
